A nossa \u00eanfase principal tem sido nas doen\u00e7as malignas hematol\u00f3gicas e, especialmente, na leucemia linfobl\u00e1stica aguda de c\u00e9lulas T (T-ALL). Uma vez que queremos compreender os mecanismos atrav\u00e9s dos quais as c\u00e9lulas T malignas adquirem uma vantagem selectiva sobre as suas cong\u00e9neres saud\u00e1veis, estamos tamb\u00e9m interessados em estudar caracter\u00edsticas espec\u00edficas da biologia das c\u00e9lulas T normais (por exemplo, a forma como os tim\u00f3citos em desenvolvimento respondem a determinadas pistas extracelulares, nomeadamente a interleucina 7).<\/p>\n\n<p>A fim de compreender se os mecanismos oncog\u00e9nicos que identificamos na LLA-T se estendem a outros cancros, estudamos outras leucemias, especialmente a LLA de c\u00e9lulas B. Por \u00faltimo, inici\u00e1mos recentemente novas vias de investiga\u00e7\u00e3o sobre os tumores cerebrais e o cancro do pulm\u00e3o.<\/p>\n\n<p>De um modo geral, a nossa investiga\u00e7\u00e3o visa esclarecer o papel que as aberra\u00e7\u00f5es intr\u00ednsecas \u00e0s c\u00e9lulas e os factores microambientais podem desempenhar durante o in\u00edcio e a progress\u00e3o do tumor, a metastiza\u00e7\u00e3o e a resposta ao tratamento. O nosso objetivo \u00e9 caraterizar os mecanismos celulares e moleculares subjacentes a estes processos, identificando genes e vias de sinaliza\u00e7\u00e3o que est\u00e3o implicados na manuten\u00e7\u00e3o e expans\u00e3o do cancro em diferentes fases.<\/p>\n\n<p>Para tal, utilizamos material de pacientes, como fonte fundamental de conhecimentos sobre a doen\u00e7a, e integramos diferentes t\u00e9cnicas bioqu\u00edmicas, celulares e de biologia molecular com modelos in vitro e in vivo adequados &#8211; permitindo uma aprecia\u00e7\u00e3o global das nuances moleculares, celulares e sist\u00e9micas associadas ao cancro. Em \u00faltima an\u00e1lise, a nossa investiga\u00e7\u00e3o procura identificar e caraterizar biomarcadores e alvos moleculares cruciais para o desenvolvimento de terapias novas e mais selectivas contra o cancro. O nosso objetivo final \u00e9 ir desde as descobertas fundamentais at\u00e9 ao desenvolvimento de novas estrat\u00e9gias terap\u00eauticas.<\/p>\n\n<p>Algumas das nossas actuais linhas de investiga\u00e7\u00e3o e esfor\u00e7os de transfer\u00eancia de tecnologia incluem:<\/p>\n\n<ol>\n<li>A carateriza\u00e7\u00e3o aprofundada da sinaliza\u00e7\u00e3o IL-7\/IL-7R e a forma como contribui para o desenvolvimento da leucemia e de outros cancros.<\/li>\n\n\n\n<li>A an\u00e1lise dos mecanismos pelos quais a intera\u00e7\u00e3o entre a CK2 e a PTEN\/PI3K modula a LLA-T.<\/li>\n\n\n\n<li>O desenvolvimento aplicado e a explora\u00e7\u00e3o de novas terapias contra T-ALL (respons\u00e1vel: Rita Fragoso).<\/li>\n\n\n\n<li>Identifica\u00e7\u00e3o dos principais determinantes moleculares das met\u00e1stases cerebrais (respons\u00e1vel: Cl\u00e1udia
