Os tel\u00f3meros s\u00e3o estruturas nucleoproteicas heterocrom\u00e1ticas que “tapam” a extremidade dos cromossomas eucari\u00f3ticos lineares. Os tel\u00f3meros s\u00e3o essenciais para proteger as extremidades dos cromossomas e para impor um tempo de vida finito \u00e0s c\u00e9lulas e aos tecidos. Os tel\u00f3meros disfuncionais causam uma instabilidade cromoss\u00f3mica grave, desencadeando assim cascatas de reac\u00e7\u00f5es celulares que s\u00e3o caracter\u00edsticas comuns de doen\u00e7as humanas como o cancro e o envelhecimento prematuro.<\/p>\n\n
Utilizamos a biologia molecular, a biologia celular e a bioqu\u00edmica para compreender como os tel\u00f3meros executam as suas fun\u00e7\u00f5es protectoras e como as disfun\u00e7\u00f5es telom\u00e9ricas est\u00e3o mecanicamente ligadas a condi\u00e7\u00f5es patol\u00f3gicas. Em particular, estudamos a forma como o RNA longo n\u00e3o-codificante telom\u00e9rico TERRA participa na manuten\u00e7\u00e3o da estrutura e fun\u00e7\u00f5es adequadas dos tel\u00f3meros, tanto em c\u00e9lulas humanas normais como doentes.<\/p>\n\n
Atualmente, estamos a explorar a forma como o TERRA apoia o alongamento dos tel\u00f3meros nas c\u00e9lulas cancer\u00edgenas, quer promovendo a atividade da transcriptase reversa especializada telomerase, quer desencadeando a s\u00edntese de ADN telom\u00e9rico mediada por recombina\u00e7\u00e3o hom\u00f3loga. Estamos tamb\u00e9m a investigar as consequ\u00eancias da perturba\u00e7\u00e3o da transcri\u00e7\u00e3o e localiza\u00e7\u00e3o do TERRA na integridade dos tel\u00f3meros e na fisiologia celular.<\/p>\n\n
