O nosso laborat\u00f3rio demonstrou que os parasitas se adaptam metabolicamente ao microambiente dos tecidos e reduzem a sua taxa de replica\u00e7\u00e3o, o que pode ter implica\u00e7\u00f5es importantes na patog\u00e9nese, no diagn\u00f3stico e nas estrat\u00e9gias terap\u00eauticas. Atualmente, a nossa investiga\u00e7\u00e3o centra-se na elucida\u00e7\u00e3o dos mecanismos fundamentais que permitem ao parasita atravessar a vasculatura, colonizar tecidos e adaptar-se ao ambiente. Al\u00e9m disso, estamos a explorar as graves consequ\u00eancias da coloniza\u00e7\u00e3o do tecido adiposo no hospedeiro, onde os animais podem perder at\u00e9 60% da sua massa gorda em apenas oito dias devido \u00e0 ativa\u00e7\u00e3o da lip\u00f3lise. O nosso objetivo \u00e9 identificar os sinais que desencadeiam esta perda dr\u00e1stica, compreender a remodela\u00e7\u00e3o do tecido adiposo durante a infe\u00e7\u00e3o e avaliar o impacto fisiol\u00f3gico global no organismo infetado.<\/p>\n<h4 style=\"font-weight: 400\">MUDAN\u00c7A DE PELE.<\/h4>\n<p>Para persistir no hospedeiro, o <em>Trypanosoma brucei<\/em> adota uma sofisticada estrat\u00e9gia de evas\u00e3o imunit\u00e1ria, alterando periodicamente o seu revestimento antig\u00e9nico, composto por glicoprote\u00ednas de superf\u00edcie variante (<em>Variant Surface Glycoproteins<\/em>, VSG). A nossa investiga\u00e7\u00e3o tem-se debru\u00e7ado sobre a complexa maquinaria molecular que regula esta varia\u00e7\u00e3o antig\u00e9nica, com especial enfoque nos processos de remodela\u00e7\u00e3o da cromatina e nas modifica\u00e7\u00f5es do RNA, incluindo N6-metiladenosina (m6A).
